Isolated sampled for DNA extraction (Cetyltrimethylammonium bromide method) and subsequent whole genome sequencing using an Oxford Nanopore Technologies (ONT) MinION Mk1B device and R10.4 flow cells (FLO-MIN114) and the ONT Rapid Barcoding Kit with an added Long Fragment Buffer. Sequencing quality was high (Q20+ 91.7%, Q30+ 85.7%), and average sequencing depth 195. Initial screening found that this sequence was not compatible with Mtb. This sequence was then analysed with the Genome Taxonomy database (GTDB, release 226) and uploaded to the Genome-to-Genome distance calculator on the Type (Strain) Genome Server (TYGS). In both cases, the sequence was assigned as a new species with placement in the genus Nocardia.
