Abstract Sil.va.ni.grel'la. N.L. fem. dim. n. Silvanigrella named after Silva nigra the Latin geographic name of the Schwarzwald (Black Forest) mountains located in the south‐west of Germany. The genus Silvanigrella accommodates heterotrophic freshwater bacteria isolated from the water column of circumneutral or slightly acidic (pH 5–7) lakes and ponds with low ion concentrations (conductivity < 30 μS/cm). So far, only the two species Silvanigrella aquatica and Silvanigrella paludirubra have been described. Cells of the type strains have in common red pigmentation, motility, and pleomorphic morphology. In both species, the cell morphology ranges from rod‐shaped cells to filamentous forms, the cell size is variable in length and width. The type strain of S. aquatica in addition forms, densely coiled spirals. The type strains of both species grow at temperatures above 30°C, but only the type strain of S. paludirubra grows at temperatures below 10°C. Growth occurred up to supplementary NaCl concentrations of 1.0–1.1% (w/v). Both type strains utilized various organic substrates including carbohydrates, amino acids, and short‐chain fatty acids; however, the substrate spectra of the two strains differed. The most abundant fatty acid was iso‐C 15:0 for both type strains, contributing 33% to the total fatty acid content. The major respiratory quinone of S. paludirubra is menaquinone MK‐8, while the quinones of S. aquatica remained unidentified. The type strains of S. aquatica and S. paludirubra were both characterized by medium‐sized genomes (3.5 and 3.9 Mb, respectively) with low G + C content (32.6 and 29.3 mol%, respectively). The genus Candidatus Nanohalobium was established based on a genome sequence found after the enrichment, cultivation, and characterization of a binary association with its chitinolytic host, obtained from a crystallizer pond brine collected in the marine solar saltern Saline della Laguna of Trapani, Italy. Ca . Nanohalobium is phylogenetically affiliated with members of the Ca . Nanohaloarchaeota ( Candidatus Haloredivivus, Candidatus Nanosalina, and Candidatus Nanosalinicola) of the archaeal DPANN superphylum. Cultivation experiments and analysis of the 0.97 Mb genome of Ca . Nanohalobium constans LC1Nh revealed that the organism has an organoheterotrophic, sugar‐fermenting lifestyle, lacking key anabolic machinery and any respiratory complexes. Additionally, the cultivation experiments revealed a remarkable mutualism of a nanohaloarchaeon–haloarchaeon association, namely the nanohaloarchaeon's ability to hydrolyze glycogen and starch to glucose, enabling the growth of Halomicrobium sp. LC1Hm in the absence of chitin. The low‐median isoelectric point for the predicted proteins suggests a “salt‐in” strategy used by the nanohaloarchaeon for osmotic balance. 16S rRNA gene sequences affiliated with Ca . Abstract Ni.tro.so.pe.la'gi.cus. N.L. masc. adj. nitrosus nitrous; L. masc. adj. pelagicus of the open sea; N.L. masc. n. Nitrosopelagicus a nitrite producer of the open sea. The genus Candidatus Nitrosopelagicus is composed of marine ammonia‐oxidizing archaea, almost all of which are uncultivated. They are found in shallow waters (<200 m depth) of the oligotrophic tropical, subtropical, and temperate open oceans. A single species has been described, Candidatus Nitrosopelagicus brevis, which couples growth with the oxidation of ammonia to nitrite. One described strain ( Ca . N. brevis U25) is capable of hydrolyzing urea to supply the ammonia required for growth. Cells are obligately aerobic, small, and nonmotile rods. They grow free‐living, not associated with surfaces. No growth enhancement has been observed in the presence of organic carbon compounds; a reduction in the lag phase of growth has been observed with addition of α‐ketoacids, potentially serving as radical oxygen scavengers. Organisms are mesophilic and obligately marine. Lipid composition distinguishes Ca . Nitrosopelagicus from Nitrosopumilus using glycerol dibiphytanyl glycerol tetraether (GDGT) distributions, with lower amounts of GDGT‐2 and higher amounts of GDGT‐0 and GDGT‐1 found in Ca . Nitrosopelagicus. Abundance of Ca . Nitrospelagicus‐like organisms is correlated with nitrification rates, suggesting that these organisms are the dominant ammonia oxidizers in epipelagic and upper mesopelagic environments of the low‐ and mid‐latitude open oceans. Halectosymbiota an ectosymbiont from salt water organisms. The genus Candidatus Halectosymbiota corrig., originally described as Ca . Haloectosymbiotes, was established based on a 16S rRNA taxonomic marker gene sequence of the only identified species in this genus, Candidatus Halectosymbiota riaformosensis. Ca . Halectosymbiota riaformosensis was observed to live as ectosymbiont on a halophile scuticociliate, which was isolated from a thalassohaline solar saltern pond (12% salinity) in Faro, Portugal. Comparisons of 16S rRNA gene sequences showed that Ca . Halectosymbiota is phylogenetically affiliated to the genera Halolamina (sequence similarity to H. pelagica : 87%) and Halobaculum (sequence similarity to H. magnesiiphilum 85%). Based on scanning electron microscopy micrographs and fluorescence in situ hybridization, cells are rod‐shaped and approximately 1.8 × 0.6 μm in size. Cells are densely covering the surface of the host, with approximately 0.52 epibiotic cells per μm 2 . Free‐living forms were not observed. Candidatus Halectosymbiota riaformosensis is sensitive to anisomycin. Known habitats are aquatic hypersaline environments. Type species : Not applicable. Cells are nonmotile, sheathed rods with flat ends. Gram‐stain negative. Lipids contain myo ‐inositol, ethanolamine, and galactose as the polar head groups. Species are oxygen‐tolerant anaerobe. However, growth and methanogenesis can only occur in strictly anaerobic and highly reducing conditions. Energy is obtained by splitting acetate into methane and CO 2 . When cocultured with electrogenic Geobacter spp., CO 2 is reduced to methane via direct interspecies electron transfer (DIET). Optimum growth pH range is 7.0–7.8. Mesophilic and thermophilic. Optimum growth temperatures are 34–37°C for mesophilic strains and 55–60°C for thermophilic strains. They occupy a wide range of anoxic habitats including anaerobic river mud, paddy field soil, hot springs, thermal lakes, thermophilic, and mesophilic anaerobic wastewater digesters treating domestic wastes; granular sludge, anaerobic fixed‐bed reactors, and other types of anaerobic systems. Genome sequences have been determined for strains of all three of the described species in the order, and the genome size ranges from 1.9 to 3.0 Mb. The order Methanotrichales consists of one family Methanotrichaceae . Although all three described species are currently classified in a single genus Methanothrix , phylogenomic analyses suggest that two species should be transferred to novel genera. Abstract Phy.to.plas‐ma. Gr. masc. n. phytos a plant; N.L. neut. n. Phytoplasma a form from a plant. Phytoplasmas are wall‐less, nutritionally fastidious, and phytopathogenic prokaryotes 0.2–0.8 μm in diameter that morphologically resemble nonhelical members of the class Mollicutes . Sequencing of nearly full‐length PCR‐amplified 16S rRNA genes, combined with earlier studies, provided the first comprehensive phylogeny of the organisms and showed that they constitute a unique, monophyletic clade within the Mollicutes . These organisms are most closely related to members of the genus Acholeplasma within the Anaeroplasma clade as defined by Weisburg et al. Despite decades of efforts and recent progress, sustained pure culture in cell‐free media has not yet been demonstrated for any phytoplasma. Their genome sizes have been estimated to range from 530 to 1,350 kb, and the DNA G + C content is about 23–30 mol%. The presence of a characteristic oligonucleotide sequence in the 16S rRNA gene, CAA GAY BAT KAT GTK TAG CYG GDC T, and standard codon usage indicate that phytoplasmas represent a distinct genus‐level taxon for which the name ‘ Candidatus Phytoplasma’ has been adopted. Thermohalobacteraceae , the Thermohalobacter family. Free‐living bacteria that mainly occur in anoxic saline environments. Cells are not pigmented and have a Gram‐positive type of cell wall. Gram‐stain reaction can be negative or positive. Typically slender rod‐shaped cells that are motile by flagella. Several strains form heat‐resistant spores. Mainly euryhaline, moderately thermophilic and halophilic. Members of the family are strictly anaerobic bacteria that utilize carbohydrates or peptides by fermentation. Cellular fatty acid patterns are dominated by iso ‐C 15:0 and iso ‐C 15:0 DMA. The family currently accommodates members of the genera Thermohalobacter , Caldisalinibacter , Caloranaerobacter , Brassicibacter , Sporosalibacterium , and Clostridiisalibacter , which were previously affiliated with the family Clostridiaceae . The affiliation of novel species to this family depends on the phylogenetic position, which should be determined on the basis of comparative sequence analyses of 16S rRNA genes or genomes. Forms short (∼5–25 μm) to long (>150 μm) flexible chains of cells within the sheath. Nonmotile. Gram‐stain‐negative. Lipids contain myo ‐inositol, ethanolamine, and galactose as the polar head groups. Oxygen‐tolerant anaerobe. Organotrophic, splitting acetate into methane and CO 2 for energy generation. Growth factors such as vitamins are stimulatory. Yeast extract is required, stimulatory or inhibitory, depending on the strain. NaCl is not required for growth. Optimal temperatures range from 34 to 37°C for mesophilic strains and 55 to 60°C for thermophilic strains; optimal pH range is 7.0–7.8. Gas vacuoles are generally found in thermophilic strains. The genus Candidatus Haloredivivus was established based on a sequence of a single‐cell‐amplified genome recovered from a solar saltern pond of intermediate salinity in Spain. Ca . Haloredivivus is phylogenetically affiliated with members of the Candidatus Nanohaloarchaeota branch ( Candidatus Nanosalina, Candidatus Nanosalinicola corrig., and Candidatus Nanopetraeus corrig.) of the archaeal DPANN superphylum. Analysis of the ∼1.2 Mb genome of Ca . Haloredivivus with 42.1% G + C suggests that the organism leads a photoheterotrophic lifestyle and may be able to degrade different polysaccharides. The low median isoelectric point for the predicted proteins suggests a “salt‐in” strategy for osmotic balance. Gene sequences affiliated with Ca .
